Sample GSM517351 Query DataSets for GSM517351
Status Public on Mar 01, 2011
Title Female rats vs male rats rep3
Sample type RNA

Channel 1
Source name Liver from female Sprague-Dawley rat
Organism Rattus norvegicus
Characteristics sex: Female
age: 7-weeks old
tissue: Liver
strain: Sprague-Dawley
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from frozen livers using Trizol following manufacturer's instructions
Label Cy5
Label protocol 5µg of total RNA was used for a first strand cDNA synthesis of each sample, labelled with either Cy3- or Cy5-dCTP. The microarray experiment was performed using the Pronto! Plus systems according to the manufacturer´s instructions (Corning and Promega).

Channel 2
Source name Liver from male Sprague-Dawley rat
Organism Rattus norvegicus
Characteristics sex: Male
age: 7-weeks old
tissue: Liver
strain: Sprague-Dawley
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from frozen livers using Trizol following manufacturer's instructions
Label Cy3
Label protocol 5µg of total RNA was used for a first strand cDNA synthesis of each sample, labelled with either Cy3- or Cy5-dCTP. The microarray experiment was performed using the Pronto! Plus systems according to the manufacturer´s instructions (Corning and Promega).

Hybridization protocol Cy3- and Cy5 labelled samples were purifyed and mixed before addition of a hybridization buffer and added to respectively array for 18h of hybridization. After hybridization, the slides were washed multiple times, all according to the Pronto! Plus systems and the manufacturer´s instructions (Corning and Promega).
Scan protocol Scanning was performed using a Genepix 4200A scanner (Axon).
Images were quantified using Genepix 6.0 software (Axon).
Description Biological replicate 3 of 4.
Data processing Background was subtracted using the in-house Easy Filter software. The data was normalized by the LOWESS method using the R language. The normalized data was finally statistically analyzed using SAM.
