The flies were raised for three more days to recover from CO2 exposure and collected using dry ice. RNA was prepared using TRI Reagent (SIGMA) according to the manufacturer’s protocol 3' RNA-seq library was performed based on Guttman’s lab "RNAtag Seq protocol" (Shishkin, A. A. et al. Simultaneous generation of many RNA-seq libraries in a single reaction. Nat. Methods 12, 323–325 (2015).)

Sequencing was done using Next seq 500 high output kit V2 75 cycles (illumina) on illumina’s NextSeq 500 system RNA-seq reads were aligned by STAR to the full genome sequences of Drosophila Melanogaster (UCSC version dm3) Reads were counted per annotated gene using ESET (End Sequencing analysis Toolkit) Genome_build: dm3 Supplementary_files_format_and_content: Table with raw gene counts for every gene and every sample
