Equal mass of (1) synovial tissue and (2) articular cartilage and subchondral bone were stored in RNAlater Stabilization Solution (AM7020, Invitrogen) at -80°C until further processing. Samples were homogenized with the Tissuelyser 2 from Qiagen (Redwood City, CA) before total RNA from the was extracted using Qiagen RNeasy Kit for purification of total RNA according to the manufacturer's instructions. mRNA expression analysis with the NanoString nCounter® Mouse Inflammation v2 Panel was performed according to the manufacturer's protocol (NanoString Technologies Inc., Seattle, WA).
