Treatment protocol Before androgen treatment, the LNCaP cells were cultivated with Phenol Red -free RPMI 1640 (GIBCO/ BRL) supplemented with 10% charcoal-dextran-stripped FBS for 5 days. Then the cells were induced with androgen dihydrotestosterone (DHT), at ultimate concentration 10 nM, which has been proven by pre-experiment to be optimum for LNCaP cell growth (data not shown). The cell response to androgen treatment was analyzed at ten time points — 0h, 20min, 40min, 1h, 2 h, 4 h, 8 h, 16h, 24 h and 48 h, where ‘0h’ refers to cell status before androgen action.
Growth protocol The androgen-sensitive human prostate cancer cell line LNCaP was obtained from ATCC and maintained in RPMI 1640 supplemented with 10% fetal bovine serum (FBS) and penicillin (100 units/ml)-streptomycin (100g/ml) at 37 ℃ in a humidified 5% CO2 cell incubator.
Extracted molecule total RNA
Extraction protocol For cell samples at each time point, the total RNA was extracted and purified using the RNeasy Mini kit following the manufacturer’s instructions. The isolated RNA was stored at -70℃ for the Illumina microarrays.
