Extracted molecule total RNA
Extraction protocol To isolate plasma miRNAs, we used plasma from familial hypercholesterolemia patients and miRNeasy Serum/Plasma Advanced Kit (Qiagen Inc), according to manufacturer’s instructions.
Label Biotin
Label protocol We used Affymetrix FlashTag Biotin HSR RNA Labeling Kit (Affymetrix, Rev3) to label 8 ul of total RNA isolated with miRNeasy Serum/Plasma Advanced Kit (Qiagen Inc) from each sample, according to manufacturers instructions. RNA quantitation was not considered due to low levels of total RNA isolated from plasma.
 
Hybridization protocol Hybridization was performed on Affymetrix miRNA 4.0 arrays and cartridges were incubated at 48°C with 60 rpm rotation for 48 h using Affymetrix Hybridization Oven 600. After, each array was washed and stained using the FS450_002 fluidics script in Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned by using the GeneChip scanner 3000 7G System
Description Circualting miRNA data in absence of coronary calcium score
Data processing The data were explored using the Transcriptome Analysis Console software (TAC 4.0) for data summarization and normalization by RMA+DABG procedure (implemented in TAC 4.0 software).
