Treatment protocol |
Cells were treated for 3 days with 1, 10, 50ug/mL DEHP (in 0.1% DMSO) or with vehicle control (0.1% DMSO).

Growth protocol |
Cardiomyocytes isolated from 2-day-old Sprague-Dawley rat pups using an enzymatic digestion protocol. Cells were plated on laminated coverslips. Cells were cultured for 3 days prior to treatment.

Extraction protocol |
RNA was extracted from cell monolayers using TRIzol reagent (Invitrogen) and total RNA was purified using RNeasy columns (Qiagen) according to manufacturer's protocols.

Biotinylated cRNA was generated according to the standard Affymetrix protocol from 5 ug of total RNA (GeneChip Expression Analysis Technical Manual; Affymetrix).

Hybridization protocol |
Following fragmentation, 15 ug of cRNA was hybridized for 16 hr at 45C on the Rat 1.0 ST arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the Affymetrix GeneChip Scanner 3000.

Gene expression data from cardiomyocyte monolayers treated with vehicle control (DMSO), 3 days.

Data processing |
Data was exported from Affymetrix software (CEL files). Analysis was conducted using GeneSpring GX10. The GeneSpring software performed a baseline transformation, which takes the median of all samples. Quantile normalization (RMA) and summarization were then performed.
