Hybridization protocol |
After prehybridization, the chip was hybridized to small RNAs labeled with Cy5 (Mirus Bio, Madison, WI) overnight at 37°C. Posthybridization washes were done sequentially once each with 6XSSPET and 3XSSPET and twice with 0.5XSSPET.

Scan protocol |
Hybridized chip was scanned using a Genepix 4000B (Molecular Devices, Sunnyvale, CA), and data were extracted using software provided by CombiMatrix (Mukilteo, WA).

Description |
small RNA A total of 12 hybridization intensities in three biological replicates per a species were obtained, and log values of hybridization intensities were used for normalization and statistical analysis.

Data processing |
If the hybridization intensities of a miRNA were not significantly higher than the background signals, the miRNA was considered to be not expressed.
