Sample GSM5553491 Query DataSets for GSM5553491
Status Public on Jan 25, 2023
Title D8_CK138: D8 TTNtv/+ AMG2591713 treated CMW
Sample type SRA

Source name cardiac microwire culture
Organism Homo sapiens
Characteristics tissue: cardiac microwire culture
treatment: AMG2591713
genotype: TTNtv/+
Treatment protocol 14 days
Growth protocol 300 μl of CMW maintenance mediumcontaining DMEM/F12, 2% fetal bovine serum, 10 μg/ml insulin and 1xantibiotic/antimycotic (all from ThermoFisher)was gently added to each well for continuous culturing. Medium is changed every 2-3 days by very slowly removing halfof medium from the side of the well at half-way heightand through the drop-wise addition of halfof fresh CMW maintenance medium.
Extracted molecule polyA RNA
Extraction protocol cardiac microwires were dissociated into a single cell suspension and encapsulated with the 10x Chromium
Samples were processed using the 10x Genomics 3'v3 kit

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina NovaSeq 6000

Data processing reads were aligned and count tables generated using cellranger v 4.0.0
Genome_build: GRCh38 (hg38)
Supplementary_files_format_and_content: barcode tsv files include cellbarcodes, features tsv files include gene identifiers, matrix files are the cell by gene count table encoded as a sparse matrix in the Market Exchange Format (MEX)
