Treatment protocol |
100,000 MDDC were plated in 96 well U bottom plate in the presence of 5 micrograms/ml of polybrene. MDDC were infected with HIVGFP(G) and SIVVLP(G) supernatants that were generated by transient transfection of 293T cells. Celles were harvested 48 hours after infection.

Growth protocol |
Human CD14+ monocytes were isolated from buffy coats of adult peripheral blood using a Ficoll gradient and two sequential purifications with anti-CD14 magnetic beads (Miltenyi). Purified monocytes were cultured in the presence of GM-CSF and IL-4 for 4 days in RPMI media containing 10% fetal bovine serum and antibiotics.
