Treatment protocol Cells were infected with retroviruses carrying carrying Gata4, Hand2, Mef2C, and Tbx5 and cultured for various time period.
Growth protocol Cells were cultured in medium composed of DMEM/199 (4:1) (Gibco), 10% FBS (Gemini), 5% horse serum (Gemini), antibiotics (Gibco), 1X non-essential amino acids (Gibco), 1X essential amino acids (Gibco), 1 B-27 (Gibco), 1 insulin–selenium–transferin (Gibco), 1 MEM vitamin solution (Gibco) and 1X sodium pyruvate (Gibco). The medium was changed every 2–3 days until cells were harvested for RNA extraction.
Extracted molecule total RNA
Extraction protocol Total RNA were extracted using TRizol reagent (Ambion) and the RNeasy Plus Universal Mini Kit (Qiagen).
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5.5 ug total RNA (GeneChip WT Plus Reagent Kit, 2013, Affymetrix).
 
Hybridization protocol Following fragmentation, 5 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Cartridge Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the GeneChip Scanner 3000 7G.
Description Total RNA extracted from mouse embryonic fibroblasts infected with retroviruses carrying Gata4, Hand2, Mef2C, and Tbx5 day 9 post-infection.
Data processing The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
Normalized data file includes MAS5.0 signal intensity
