|
Source name |
liver from casein fed rats
|
Organism |
Rattus norvegicus |
Characteristics |
tissue: liver diet: casein age: 20 weeks strain: Diet-induced obese Sprague-Dawley gender: male
|
Treatment protocol |
Rats were over-anaesthetized with isoflurane and killed via aortic cut. Liver was excised and submerged in liquid nitrogen immediately. Liver was stored at -80˚C until analysis.
|
Growth protocol |
12 week old rats were provided one of 3 ad libitium diets for 8 weeks. Diets were prepared with 10% (by kcal) of either casein, leucine, or dairy protein.
|
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted from liver using Trizol Reagent (Invitrogen, Carlsbad, CA) and purified using the Qiagen RNeasy Miniprep (Qiagen, Mississauga, ON, Canada). RNA was quantified using Ribogreen and RT performed using the Omniscript RT Kit as per manufacturers directions (Qiagen, Mississauga, ON, Canada).
