The harvested-mycelia (WT, ΔrgsA and ΔrgsE) were frozen in liquid nitrogen, and were lyophilized for 1day. The RNA of WT, ΔrgsA and ΔrgsE strains was extracted by TRIzol reagent according to the manufacturer’s instruction. RNA libraries were prepared for sequencing using standard Illumina protocols.

Illumina Casava1.7 software used for basecalling Raw sequencing data was first filtered by Trimmomatic (version 0.36), then mapped to the reference genome of aspergillus flavus using STRA software (version 2.5.3a). Reads mapped to the exon regions of each gene were counted by featureCounts (Subread-1.5.1; Bioconductor) and then RPKMs were calculated. Genes differentially expressed between groups were identified using the edgeR package (version 3.12.1).
