Different doses of hnrnpaba MO reagent (3ng and 6 ng) were microinjected into each zebrafish embryos at the 1-cell stage. A standard control MO was used as controls. The injected embryos were raised at 28.5℃ and harvested at 24 hpf for RNA extraction.

30 pooled embryos at 24 hpf from different experiment were snap-frozen in liquid nitrogen and stored at -80°C. Total RNA of each group was extracted with Trizol (Invitrogen) and assessed with Agilent 2100 BioAnalyzer (Agilent Technologies, Santa Clara, CA, USA) and Qubit Fluorometer (Invitrogen).
Library construction and sequencing were performed on a BGISEQ-500 by CapitalBio Technology (www.capitalbiotech.com, Beijin, China). Clean-tags were mapped to the reference genome and genes available at the Zebrafish Genome.

For gene expression analysis, the matched reads were calculated and then normalized to RPKM using RESM software
The significance of the differential expression of genes was defined by the bioinformatics service of BGI according to the combination of the absolute value of log2 Ratio ≥1 and FDR ≤ 0.001.
Library construction and sequencing were performed on a BGISEQ-500.
