Animals were anaesthetized using 0.75 ml/kg Hypnorm s.c. (fentanyl/fluanisone; Jansen Pharma, Birkeroed, Denmark) and 4 mg/kg Midazolam s.c. (dormicum; La Roche, Basel, Switzerland) and placed on a heated pad. A midline laparotomy was performed and the hepatoduodenal ligament was mobilised. Liver biopsies were sampled from the right lobe and immediately snap frozen in liquid nitrogen, and stored at -80°C until analysis.

Male Wistar rats, weighing 300–350 g, (M&B Taconic, Eiby, Denmark) were housed in standard animal laboratories with the temperature maintained at 23°C and artificial 12-hour light-dark cycles, and with free access to food and water.

Biopsies were homogenized in RLT plus buffer on Qiagen Tissuelyser. Total RNA was extracted on the QIAsymphony SP using the QIAsymphony RNA extraction kit.

150 ng of total RNA was used as target preparation in the starting material for the cDNA Syntesis Kit and WT Terminal Labeling Kit (affymetrix).

The target was loaded onto the GeneChip and hybridised for 16 h. The arrays were washed and stained in the Affymetrix Fluidics Station.

GeneChips were scanned using Affymetrix 7G GeneChip Scanner.

Raw image files from the quantitative scannings were analysed by Affymetrix Gene Expression Analysis Software and data were normalized using the RMA in Affymetrix Expression Console.
