On day 6 of CTX-induced skeletal muscle regeneration, TA muscles of both hindlimb legs were dissected, digested, and subjected to flow cytometry. After exclusion of doublets and dead cells, myogenic progenitor cells were sorted based on the expression of alpa7 integrin and the exclusion of CD45/CD31/sca1. Subsequently, single cell suspension was generated using Chromium. Chromium Single Cell 3' Reagent Kits User Guide (v3.1 Chemistry).

Data processing |
Mapping was performed by cellranger. Cellbender was used to remove empty droplets and ambient RNA. scDblFinder was used to exclude potential doublets. Downstream analysis was performed by Seurat. Assembly: GRCm39 Supplementary files format and content: Raw count matrices from Cell Ranger. barcodes.tsv, features.tsv, matrix.mtx files.
