Growth protocol |
Normal human atrial cardiac fibroblasts, normal human ventricular cardiac fibroblasts, normal human dermal fibroblasts and bone marrow-derived mesenchymal stem cells were cultured in commercially available growth media (lonza): FGM-2 for normal human dermal fibroblasts, FGM-3 for normal human atrial cardiac fibroblasts and normal human ventricular cardiac fibroblasts, MSCGM for bone marrow-derived mesenchymal stem cells. These primary cells were used at passages 5-7. iPS cell-derived fibroblasts were prepared from 201B7 or 253G1 cell lines.

Extraction protocol |
Total RNA was extracted from cells using an RNeasy Plus Mini Kit (Qiagen, Hilden, Germany), according to manufacturer's instructions. RNA quantity and quality were determined using a Nanodrop ND-1000 spectrophotometer (Thermo Fisher Scientific Inc.) and Agilent Bioanalyzer (Agilent Technologies, Santa Clara, CA), as recommended by the manufacturers.

0.6 ug of Cy3-labelled cRNA was fragmented at 60°C for 30 minutes in a Agilent fragmentation buffer and was hybridized at 65°C for 17 hr using Gene Expression Hybridization Kit.

Scan protocol |
Microarray slides were scanned at 3 μm resolution using an Agilent SureScan (G4900DA).

Data processing |
The scanned images were analyzed with Feature Extraction Software 10.7.3.1(Agilent) using default parameters to obtain background subtracted and spatially detrended Processed Signal intensities. Features flagged in Feature Extraction as Feature Non-uniform outliers were excluded.
