Testes from pre-pubertal Holstein breed cattle and mixed breed pigs were collected by castration. For all samples, single-cell suspensions of testicular parenchyma were generated as described previously (Oatley and Brinster, 2006; Oatley et al., 2016). Live cells (~8000 per sample) were then loaded into a Chromium Controller (10X Genomics, Inc.), and cDNA libraries were generated using v2 chemistry kits (10X Genomics, Inc.). Libraries were pooled at proportions netting equal read depth and sequenced in a single lane on an Illumina HiSeq 4000 (Genomics and Cell Characterization Core Facility, University of Oregon).
