Treatment protocol |
Spontaneous hypertension rats were treated with empaglifozin by gavage at the dose of 10 mg/kg/d or saline vehicle control daily for a period of 30 days.

Growth protocol |
Spontaneous hypertension rats were housed in groups of 3-4 rats/cage in a controlled environment (22 ± 2°C, 12-h/12-h light/dark cycle with lights on at 7:00 am) and ad libitum access to water and standard rat chow.

Total RNA was extracted from the rat tissues using Trizol (Invitrogen, Carlsbad, CA, USA) according to the manufacturer’s instructions.1.3 ug of total RNA was used for the construction of sequencing libraries. the final library was generated by heated denaturing and circularizing the above double stranded PCR products using the splint oligo sequence, then followed by formatting the single strand circle DNA (ssCir DNA). DNA nanoballs (DNBs) were generated by library amplification and loaded into the patterned nanoarray and single end 50 bases reads on BGIseq500 platform (BGI-Shenzhen, China)

SOAPnuke (v1.5.2) Sequenced reads were trimmed for adaptor sequence by SOAPnuke (v1.5.2), afterwards clean reads were obtained and stored in FASTQ format. Clean reads were mapped to rattus norvegicus genome using HISAT2(v2.0.4) Bowtie2 (v2.2.5) was applied to align the clean reads to rattus norvegicus genome Sequenced Fragments Per Kilobase of exon per Million fragments mapped to rattus norvegicus genome (FPKM) were calculated using RESEM(v1.2.12). Assembly: Rattus_norvegicus NCBI GCF_000001895.5_Rnor_6.0 Supplementary files format and content: Excel files include FPKM values for each sample.
