Sample GSM642866 Query DataSets for GSM642866
Status Public on Dec 21, 2010
Title 3134-DS8801-DEX-4-hrs
Sample type RNA

Source name mouse mammary adenocacinoma tumor, dexamethasone 4 hours
Organism Mus musculus
Characteristics cell line: 3134
term: MGI:7035
strain: RIII
tissue: mouse mammary adenocacinoma tumor
gender: male
time: Dex 4 hours hour
Biomaterial provider Hager;hager-lab
Treatment protocol Dex treated: treatment_hour with 100 nM dexamethasone. Control, untreated 0 hour.
Growth protocol Cell lines were maintained in Dulbecco's Modified Eagle Medium (DMEM) (Invitrogen, Carlsbad, CA) supplemented with 10% fetal bovine serum (Gemini, Woodland, California), 2 mM L-glutamine, 1 mM sodium pyruvate, 0.1 mM non-essential amino acids, 5 mg/ml penicillin-streptomycin (Invitrogen, Carlsbad, CA) and kept at 37ÂșC incubator with 5% CO2. Cells were transferred to 10% charcoal-dextran-treated, heat-inactivated fetal bovine serum for 48 hrs prior to hormone treatment.
Extracted molecule total RNA
Extraction protocol RNA was extracted from cells either vehicle treated or treated with 100 nM hormone (dexamethasone, Dex) for 0h, 2h, 4h or 8h. 3134 and AtT-20 RNA for microarray analysis was prepared via standard manufacturer protocols (Qiagen, Valencia, Ca) using cells resuspended in Trizol reagent (Invitrogen, Carlsbad, Ca)
Label biotin
Label protocol RNA from untreated and dex treated cells were labeled with biotin-CTP using manufacturer's recommendations (Affymetrix, Santa Clara, Ca)

Hybridization protocol Hybridizations were carried out according the manufacturer (Affymetrix) protocol.
Scan protocol Affymterix Gene Chip Scanner 3000
Description The 3134 cell line was derived by transformation of C127, originally isolated from a mammary adenocacinoma tumor of the RIII mouse. Dexamethasone treated, 4 hours.
Data processing Probe-level CEL files were processed through Affymetrix Expression Console using RMA summarization and median normalization methods.
