Total RNA was isolated from tissues with mirVana RNA Isolation labeling kit (Ambion Inc) according to the manufacturer's protocol. RNA quality and integrity were confirmed by ExperionTM Automated Electrophoresis System (BIO-RAD)

200 ng of total RNA was converted to cDNA. A single miRNA-specific oligo (MSO) was used to assay each miRNA. All MSOs were hybridized to the sample in parallel, and followed by a solid-phase primer extension step. The universal PCR step created fluorescently labeled products identifiable by their unique MSO sequence. The address sequence from each MSO was used to hybridize specific miRNA products to specific locations on the BeadArrayTM substrate for readout.

Labeled, material (15ul per array) was hybridized to the Illumina Human v2 MicroRNA expression BeadChip according to the Manufacturer's instructions (Illumina, Inc., San Diego, CA).

Data processing |
Signal intensity values were analyzed using BeadStudio version 3.1.3. We set a cut-off detection p-value of 0.01 to determine whether a probe was significantly detected. Thereafter the data of 410 probes was analyzed . The data were normalized using quantile normalization in the R language environment and log2 transformed.
