Treatment protocol None
Growth protocol Human samples were thawed and plated overnight in DMEM 20% FBS. SP and MP cells were isolated by FACS purification the next day.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from sorted human fetal muscle side population (SP) and main population (MP) cells using the RNeasy kit (QIAGEN). Total RNA (100 ng) from 22 SP and 10 MP samples were subjected to two rounds of linear amplification using a MessageAmp kit (Ambion).

Label protocol Fragmented cRNA was biotin-labeled via standard Affymetrix labeling protocol.
 
Hybridization protocol Standard Affymetrix hybridization protocol for the Affymetrix human HG-U133A GeneChip for 20 µg of cRNA.
Scan protocol Scanning is carried out on a Affymetrix GeneChip® Scanner 3000 7G scanner with autoloader. The Affymetrix GCOS v1.3 operating system controls the Model 3000 7G scanner and data acquisition functions. GCOS maintains the mediated first level data analysis and desktop data management for the entire GeneChip System.

Data processing CEL files were inputted into Bioconductor/R to derive Affymetrix Microarray Suite version 5.0 (MAS 5.0) normalized output.
