Single-cell library was prepared using the 10X Genomics Chromium Next GEM Single Cell 3’ Reagent Kits v3.1 (10X Genomics) according to the manufacturer’s instructions. Briefly, xrOvary at ag49+52 was trypsinized and applied to flow cytometry (BD, FACS Aria III). CyESC-derived germ cells were collected as FSC-W high/DRAQ7-negative/AG and/or VT-positive cells, on the other hand, mouse gonadal somatic cells were obtained as FSC-W low/DRAQ7-negative/AG-VT- population. Cy germ cells and mouse cells were mixed and the cell concentration was adjusted to 1000 cells/µL and applied to Chromium 10X Single Cell System (10X Genomics) to generate the Gel-Bead in Emulsions (GEMs). cDNA libraries were prepared using Chromium Next GEM Single Cell 3’ GEM, Library & Gel Bead Kit v3.1 (10X Genomics) following the manufacturer’s instructions.
