Cells were activated by adding BrHPP (500 nM) ad day 0.

Growth protocol |
Cells were cultured in complete RPMI 1640 medium containing 2 mM L-glutamine, 100 µg/ml streptomycin, 100 IU/ml penicillin and 1 mM sodium-pyruvate supplemented with 10% fetal calf serum and recombinant human IL-2 (100 IU/ml) added every three days.

Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 1-3 µg total RNA from human cells, amplified and labeled following the one-Cycle Target Labeling protocol (Affymetrix, Santa Clara, CA, USA).

Labeled complementary RNA (cRNA) from these samples was then fragmented and hybridized to Affymetrix GeneChip arrays HG-U133 plus 2.0.

The chips were then washed, scanned on the Affymetrix Fluidics Station 400 and analyzed with GeneChip Operating Software.

The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method.
