Growth protocol UCB-derived CD34+ cells were cultured in Dulbecco’s Modified Eagle Medium (DMEM) supplemented with 10% heat-inactivated fetal bovine serum (FBS), 1% Penicillin/Streptomycin (Pen/Strep) (MP Biomedicals), 1% Hepes buffer , SCF (50 ng/ml), IL-3 (50 ng/ml), GM-CSF (10 ng/ml) and maintained at 37°C in a humidified 5% CO2 incubator, with or without increasing doses of Denatonium Benzoate (DEN)

Extraction protocol Total RNA was isolated using a Rneasy Micro kit,according to the manufacturer’s instructions

Label protocol The fragmented cDNA is labeled by terminal deoxynucleotidyl transferase (TdT), according to the manufacturer’s instructions
 
Hybridization protocol The cDNA fragments were hybridized on cartridge array using the GeneChipTM Hybridization Oven 645 set ON at 60 rpm and 45°C.
Scan protocol The microarrays were finally scanned on a GeneChipTM Scanner 3000 System (Affymetrix).The fluorescent signals relative to hybridized probes were obtained as DAT files and were converted into CEL digital signal by Affymetrix GeneChip Command Console (AGCC) software

Data processing CEL files' raw data were normalized using Robust Multi-array Average normalization (RMA) and log-transformed using Oligo R package.Normalized data were annotated by pd.clariom.s.human
