Treatment protocol |
After surgical intervention the rabbits with Atherosclerosis were fed with 0.30% high cholesterol diet and after 8 weeks were switched to a maintance diet of 0.15% high cholesterol diet for a remainig of 12 weeks

The rabbits were housed with enrichment and had ad libitum of chow or High cholesterol diet, depending on their treatment

We sacrificed the rabbits, and surgically removed the adbominal aortas. We flushed them with saline and put cut sectios into RNAlater for storage overnight in the fridge. Then the RNAlater was removed and the tissue was tored at -20C until extraction of RNA.

Total RNA was extracted with TRIzolĀ® Reagent and reverse-transcribed into cDNA using the RT2 First Strand Kit QIAGEN. And plated out on PCR array plates. Plates were read on the Mx3000P real-time PCR system (Stratagene, CA, USA) with paramters by 1 cycle 10 minutes of 95C and then 40 cycles of 15s at 95C and 1 min at 60C
