Title |
CCl4-induced liver fibrosis for 4 weeks, biological rep3 |
Sample type |
RNA |
Source name |
Fibrosed liver, 4 weeks of CCl4 injections, biological replicate no 3
Organism |
Mus musculus |
Characteristics |
tissue: Liver gentoype: C57Bl/6N age: 14 w
Treatment protocol |
Mice were subjected to 1.6 g/kg CCl4 i.p. twice/week. Then two days after last CCl4 injection mice were sacrificed and liver was harvested
Growth protocol |
CCl4-induced fibrosis is described in https://doi.org/10.21203/rs.3.rs-2009380/v1
Extracted molecule |
total RNA |
Extraction protocol |
Total liver RNA isolation was performed using InviTrap®Spin Universal RNA Mini Kit from Stratec (1060100300, Birkenfeld, Germany) according to the manufacturer's instructions.
Label |
Biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol.
Hybridization protocol |
Hybridization (16h x 45°C) was processed according to the standard Affymetrix protocol.
Scan protocol |
Affymetrix GeneArray Scanner3000
Description |
Gene expression of mouse fibrose liver 4 weeks after CCl4 induction
Data processing |
The data were analyzed with a commercial software called JMP Genomics, version 8, from SAS. Gene expression profiling was performed using arrays of Mogene-2_0 type from Affymetrix. A Custom CDF Version 20 with Entrez based gene definitions was used to annotate the arrays. The Raw fluorescence intensity values were normalized applying quantile normalization, RMA background correction and Medianpolish Probeset Summary
