Title |
Frogs treated with artemisinin |
Sample type |
RNA |
Source name |
liver, artemisinin
Organism |
Xenopus laevis |
Characteristics |
tissue: liver gender: female treatment: artemisinin
Treatment protocol |
Arteminisin (200 mg/kg) or pregnenolone (50 mg/kg), each dissolved in DMSO, were injected intraperitoneally into three frogs on two consecutive days. The control group received in parallel two DMSO injections. All frogs were sacrificed 24 h after the second injection by decapitation, and livers were immediately frozen in liquid nitrogen.
Growth protocol |
Xenopus laevis were kept in 22°C water.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA extraction was performed with peqGOLD TriFast (Peqlab) according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA.
Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45°C on a GeneChip Xenopus laevis 2.0 Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
Description |
Gene expression data from artemisinin-treated Xenopus laevis.
Data processing |
The images were processed by GeneChip Operating Software 1.3, and the raw data were normalized by Robust Multiarray Analysis (RMA).
