| Treatment protocol |
Control guinea pigs (n=6) were given untreated water. Guinea pigs in the Dex group (n=6) were given the sGC Dexamethasone 21-phospate disodium salt in their drinking water every other day for 48 days. Weight and water measurements were taken so that Dex-treated animals would receive ~3mg/kg Dex in the drinking water. Sperm was isolated from the caput and cauda section of the epididymis following euthanasia.

12-week-old adult male Dunkin-Hartley guinea pigs were singly housed on a 12-hour light-dark cycle. Food and water were available ad libitum.

Total RNA was extracted from caput and cauda sperm using Trizol, according to the manufacturer’s recommendations, with some modifications. RNA was extracted from a total of 24 sperm samples. (Caput control n=6, Caput Dex n=6, Cauda control n=6, Cauda Dex n=6)

The data were analyzed with the Affymetrix Transcriptome Analysis Console 4.0.1(TAC) software using RMA+DMG and removing batch effects. Differential analysis was conducted between control and Dex groups for both caput and cauda microarrays and calculated using one-way ANOVA with Benjamini-Hochberg Step-UP FDR correction
