Growth protocol |
To separate CD43- from CD43+ cells, washed splenic lymphocytes (5e7) were resuspended in 450 µL FACS buffer and stained with 50 µL CD43 microbeads on ice for 30 min. Cells were washed once, resuspended in 500 µL FACS buffer and passed through an LD column in a VarioMACS separator as per manufacturer’s recommendation. CD43 microbead-labelled non-B cells were eluted from the column by applying the plunger. Purified B cells in the flow-through were incubated in RPMI-1640 supplemented with 10% (v/v) FBS, 55 µM 2-ME, 2 mM L-glutamine, penicillin and streptomycin plus 2 ng/mL IL-4 and BAFF at 37 °C in 5% CO2.
