Mice were fed with normal chow or rosiglitazone-containing normal chow (0.0075%) for 2 months.

Adult (2-6 months old) PPARgamma-tTA;tetO-H2B-GFP mice were housed in a 12:12 light:dark cycle and chow and water were provided ad libitum.

White adipose tissues (inguinal, interscapular and gonadal white adipose tissues) were digested with collagenase for 2 hours at 37 degree with gentle shaking, and stromal-vascular fraction was collected and GFP+ cells from this fraction were FACS-sorted. RNA was extracted with Trizol reagent, followed by clean-up and DNase I treatment with QIAGEN RNeasy mini kit in accordance with the prescribed protocol provided with the kit. Quality control was performed with Agilent Bioanalyser.
