Extraction protocol |
The spikelets at booting stage (Feeke 10) were harvested and frozen in liquid nitrogen for RNA isolation. 4 μg total RNA was used to prepare each sequencing sample. Libraries were constructed and sequenced by Novogene (Beijing, China) to generate 50-bp single-end reads.

Data processing |
The reads were trimmed with cutadapt to remove Illumina adapters and low-quality bases (quality score < 30). Shortstack was utilized to map the 21-nt and 24-nt clean reads. Assembly: IWGSC RefSeq v1.0 Supplementary files format and content: bw files of normalized 21nt and 24nt smRNA signals for each sRNA-seq data to display the smRNA distribution along the chromosomes
