Treatment protocol β-estradiol power (E2257,Sigma) was dissolved in DMSO with 3.7% filtered sodium hydrogen carbonate. After incubation of the final concentration of β-estradiol with 250nmol/L, cells were incubated at 37°C and 5% CO2 for 48 h before harvested.
Growth protocol HA-VSMC cells were cultured in RPMI1640 containing L-Glutamine ( C11875500BT, Gibco), and 10% FBS (10099141, Gibco), at 37℃ in a 5% CO2 incubator. Cells were seeded at a density of 2 × 105 viable cells /mL, and passaged every 3–4 days.
Extracted molecule total RNA
Extraction protocol For RNA isolation, total RNA was isolated using TRIzol reagent, in accordance with the manufacturer’s recommendations.
The mRNA sequencing libraries were generated using a TruSeq Small RNA Sample Preparation Kit, and index codes were added to attribute sequences to each sample.

Data processing Sequencing reads were trimmed using Trim Galore (parameter - Quality limit: 20)
Trimmed equencing reads were aligned to the rabbit reference genome (oryCun2) using TopHat2 (version 2.1.1).
The gene expression level was calculated by Cufflinks.
The differential expression between different groups was performed with the Cuffdiff module.
