Extraction protocol |
The total RNA of each sample was extracted using TRIZOL reagent. The extracted small RNAs were enriched and purified, and then add 3' end adaptor by ligating the 5-adenylated and 3-blocked adaptor to the 3' end of the small RNA fragment. Next, add Unique molecular identifiers (UMI) labeled Primer, and then ligate 5' end adaptor. The first strand with UMI labeled Primer was synthesized, then the second strand synthesis. Select the fragment. Take Library quantitative, pooling cyclization and Library QC. Finally, the small RNA was sequenced on DNBSEQ.

Data processing |
Reads whose first 25 nucleotides had sequencing scores less than 30 were discarded. Reads with at least 18 nt after removing the 3'-adapters were kept. The unique sequences of the remaining reads were obtained and aligned to the miRNA sequences using the BLAST algorithm with the options of ``-S 1 -m 8 -e 0.01". The abundances of miRNAs were calculated. Assembly: hg38 Supplementary files format and content: MS Excel file includes raw count and normal abundance (Reads Per Ten Million sequencing tag) of miRNAs for each sample.
