Treatment protocol Cells were cultured in 6-well plates with 3 ml media in each well. Chemical cocktails were added to the media such that final DMSO and PBS concentration was each 0.5%. Media and treatments were replaced every 3 days.
Growth protocol Fibroblasts were cultured in DMEM/F12 media supplemented with 1X anti-anti, 10% FBS, 1X beta-mercaptoethanol, and 1X non-essential amino acids (Gibco). Cells were grown at 37°C with 5% CO2 and 3% O2.
Extracted molecule polyA RNA
Extraction protocol Cells were washed 3x with PBS, and lysed in the dish with Trizol. Isolation of total RNA was performed according to manufacturer's protocol (direct-zol, Zymo Research)
Libraries were prepared with TruSeq Stranded mRNA LT Sample Prep Kit according to TruSeq Stranded mRNA Sample Preparation Guide, Part # 15031047 Rev. E
