Extraction protocol Both hindpaws were excised amputated at the ankle, and digested in a collagenase solution for 1.5-2 hours at 37°C, with agitation every 15 minutes. The resulting cell suspension and tissue remnants were filtered through a 70µm nylon filter, and the filter was rinsed to collect cells. After centrifugation and resuspension, the cells were sorted in FACS buffer (2% FBS + 0.5mM EDTA in PBS)
Library was performed according to the manufacture's instruction (single cell 3' v3 protocol, 10x Genomics)
