Treatment protocol |
AAV2-GFP or AAV2-Ezh2-Y726D was injected into the vitreous body and the optic nerve was crushed after two weeks. Three days after the optic nerve crush, retinal ganglion cells were enriched from dissociated retinal cells by fluorescence-activated cell sorting.

RNA was isolated from retinal ganglion cells using the PicoPure RNA Isolation Kit (Thermo Fisher Scientific KIT0204) following the manufacture’s manual. RNA-seq libraries were prepared using the TruSeq Stranded Total RNA Kit (Illumina).

Raw FASTQ data were mapped to the mouse reference genome (GRCm38) using STAR aligner (version 2.7.0d) with default parameters. The number of counts per gene was estimated using the “quantMode” command in STAR. Assembly: GRCm38 Supplementary files format and content: tab-delimited text file including raw count for each library
