Treatment protocol |
Co-culture assay of PBT cell lines were grown as described above and seeded to 100,000 cells/2 ml in 6-well plates. ICG-001 was added to cell cultures in concentrations 0, 5, and 10 mM for 24–72h.

Growth protocol |
PBT tumor cells lines are derived from patient with brain tumors (IRB07074) dissociated and grown in DMEM/F12 medium, supplemented with heparin, hepes, glutamax, and B27. Epidermal growth factor (EFG) and fibroblast growth factor (FGF) are added at the time of culture, as described previously(63). ICG-001 was provided by M. Kahn’s laboratory.

Hybridization protocol |
RNA was hybridized with the Codeset from the gene panel at 65°C for 16 h. The post-hybridization probe-target mixture was quantified using the nCounter Digital Analyzer, and all data analyses were performed on nSolver (NanoString Technologies).

Scanning was done using NanoString nCounter system, following manufacturer's instructions.

Data processing |
All raw data were first normalized with internal positive and negative controls to eliminate variability unrelated to the samples, then normalized to the selected housekeeping genes using Geometric Means methods. Statistically significant (p<0.05) and |fold change|≥1.5 differential expressed genes were analyzed for different cell lines and treatments.
