Total RNA was extracted from grains using M5 HiPer Plant RNeasy Complex Mini Kit (Mei5bio). 1 μg total RNA was used for the construction of sequencing libraries. The sequencing libraries construction and sequencing on the BGISEQ-500 platform were performed at Beijing Geonomics institute(BGI)

The RNA-seq clean reads were mapped to the reference genome IWGSC Refseq v1.1 using Bowtie2 Htseq-count with default parameters was used to calculate the gene read counts The gene read counts were normalized to TPM and calculated the gene expression level by Salmon. Assembly: IWGSC_Refseq_v1.1 Supplementary files format and content: text file includes read counts of genes for each sample Supplementary files format and content: text file includes TPM value of genes for each sample
