Treatment protocol |
The three rabbits without treatment were taken as the control group (NS). The other rabbits with HS lesions were randomly divided into 3 groups (three rabbits in each group): HS, drug-loaded MNs, and 5-Fu injection. Administration started from the 22nd day after skin resection, once a week for 3 consecutive weeks (on the 22nd, 29th and 36th day). For NS and HS, no treatment was given. For drug-loaded MNs, one patch of drug-loaded MNs was used in each HS lesion. For 5-Fu injection, 0.1 mL of 5-Fu solution (120 μg) was injected in each HS lesion.

RNA was harvested using Rneasy mini plus kit (Qiagen) and used for the construction of sequencing libraries. RNA-seq were prepared using SMARTER mRNA-Seq Library Prep Kit following manufacturer's protocols.

CLC Genomics Workbench v 11.0.1 Sequence reads were trimmed for adaptor sequence/low-quality sequence using CLC genomic benchwork (parameter-Quality limit: 0.05) Trimmed sequence reads were mapped to OryCun3.0 using CLC genomic benchmork (parameters-mismath cost: 2 insertion cost: 3 deletion cost: 3 length fracion: 0.8 similarity fraction: 0.8) Read count extraction and normalization were performed using CLC genomic benchwork Assembly: OryCun3.0 Supplementary files format and content: tab-delimited text files include FPKM values for each Sample.
