Treatment protocol |
After growing for 20 days, plants were separated into two batches; one irrigated with water and the other with 200 mM Na2S for 10 additional days.

Arabidopsis thaliana (accession Col-0) and the des1-1 mutant (SALK_103855 ) were grown in soil under a photoperiod of 16 h of white light (120 μE m-2 s-1) at 20ºC and 8 h dark at 18ºC.

RNA was prepared using TRIzol (Invitrogen) and the RNeasy Mini Kit (Qiagen). RNA was quantified and the RNA quality was assessed with a 2100 Bioanalyzer from Agilent Technologies

Biotinylated cRNA was prepared according to the standard Affymetrix protocol from 500 ng total RNA (GeneChip 3' IVT Express Kit).

Hybridization protocol |
Following fragmentation, 10 µg of biotinylated cRNA were hybridized to Affymetrix chips (ATH1). Each sample was added to a hybridization solution containing 100mM 2-(N-morpholino)ethanesulfonic acid, 1 M Na+, and 20mM of EDTA in the presence of 0.01% of Tween-20 to a final cRNA concentration of 0.05 µg/ml. Hybridization was performed for 16 h at 45ºC . Each microarray was washed and stained with streptavidin-phycoerythrin in a Fluidics station 450 (Affymetrix).
