Treatment protocol |
After 5 passages following thaw from -80C in cDMEM, LPS (Enzo Life Sciences) was used to stimulate the cells (100 ng / mL in cDMEM) at the 0, 30, 60, and 120 minute timepoints. After stimulation the plates (10 cm treated dishes) had the LPS containing media aspirated off the top and were immediately placed on ice. 2 washes with ice cold PBS were performed before the addition of lysis buffer.

Growth protocol |
IMM cells maintained in cDMEM. DMEM supplemented with heat inactivated 10% FBS and 0.02M HEPES in a humidified atmosphere with 5% CO2 and 37C.

Data processing |
Raw files and the RLF were imported into nSolver Analysis 4.0. Normalization was performed relative to the unstimulated 0 minute timepoint using the Normalize function provided in the software.
