All tapes collected from a single site were pooled into a single tube and underwent enzymatic digestion in a chaotropic buffer to extract genomic material from the adhesive. Nucleic acids were pricipatated and isolated using magnetic beads, automated on a KingFisher Apex instrument. Samples underwent demultiplexing and were converted to FASTQ files using BCL Convert version 2.4.0 Samples underwent splice aware mapping and alignment to human reference genome (hg38 No Alts, with decoys) using Dragen RNA version 4.0.4.
