In this work, the effect of splenectomy on liver regeneration was studied. For this purpose in the animals were reproduced splenectomy, and after 7 days 70% liver resection was performed. Animals in which the spleen was intact and only 70% liver resection was performed served as a comparison group. Also, the control group included in whom only the spleen was removed. Peripheral blood and spleen monocytes from intact animals (spleen and liver intact) and from animals in which the liver was resected (spleen intact) were isolated for transcriptome analysis.

The mouse mononuclear cell fraction was obtained from peripheral blood and spleen by gradient centrifugation. After that, the cells were subjected to immunomagnetic sorting on a manual MidiMACS™ Separator using LS Columns (Miltenyi Biotec, Bergisch Gladbach,Germany) and using Anti-CD115 MicroBeads magnetic microbeads (Miltenyi Biotec, Bergisch Gladbach, Germany) in accordance with the manufacturer's recommendations. For each sample, monocytes were obtained from 5 mice. DNBSEQ Eukaryotic Strand-specific mRNA library
