Peripheral blood was obtained from female healthy donors from different ages. Peripheral blood mononuclear cells (PBMC) were extracted using a Ficoll gradient. To separate CD19 and CD4 positive cells CD19 MicroBeads and CD4+ T cell Isolation kit II (Miltenyi Biotec) was applied following the manufacturer’s instructions. DNA was Phenol:Chloroform:Isoamylalcohol (Sigma), respectively. CD19+ B-cells were immortalized by Epstein-Barr-Virus (EBV) applying previous published protocol (Miller, G. & Lipman, M. Release of infectious Epstein-Barr virus by transformed marmoset leukocytes. Proc. Natl. Acad. Sci. U.S.A 70, 190-194 (1973)). Bisulphite modification of genomic DNA was carried out with the EZ DNA Methylation Kit (Zymo) following the manufacturer's protocol.
