Growth protocol |
The cells were cultured in serum-free LHC-8 medium (Gibco, USA) at 37°C in an atmosphere containing 95% air and 5% CO2. The culture medium was changed every 3 days. When cells reached 90% confluency, they were detached by using trypsin (Sigma-Aldrich, USA) for 5 min, and the suspension was separated by centrifugation to collect the cells. The cells were then subjected to subculturing.

DNA was extracted, purified and amplified with TruePrepTM Index Kit V2 for Illumina® (Vazyme) Libraries were analyzed for size distribution by Agilent 2100 Bioanalyzer and quantified by real-time PCR.Finally, DNA libraries were sequenced on the Illumina Novaseq platform and 150 bp paired-end reads were generated.
