Extraction protocol |
Total RNAs were extracted from frozen cell pellets using hot phenol-chloroform extraction followed by ethanol precipitation. RNA libraries were prepared from 500 ng of total RNAs using the PCR-cDNA barcoding kit from Oxford Nanopore (ONT, catalog #: SQK-PCB114.24) as per the manufacturer's instructions.

Basecalling was performed using the built in Dorado basecaller in Minknow (Dorado version: 7.4.12). Reads were then mapped to S288C reference genome using Minimap 2 (Version 2.17-r941). Read counts were generated by featureCounts and normalized using DESeq2 Assembly: R64-1-1 Supplementary files format and content: comma-separated text file includes the raw counts for each sample
