Bronchoalveolar lavage of the lungs was performed with 100mL aliguots of sterile PBS containing genamicin at 70 μg/mL(Sigma).Then lavage fluids were centrifuged at 230 ×g for 15 min, and cells were washed twice with Dulbecco’s Modified Eagle’s Medium (DMEM) containing gentamicin (50μg/mL).

| Growth protocol |
Six 28 days old healthy pigs were obtained, Three pigs were inoculated intratracheally with 2 ml of 4.5×108 colony forming units of HPS4 strain MD0322. other three pigs were intratracheally challenged with 2 ml PBS as control. All pigs were slaughtered at 28 dpi.

Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Porcine Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
