Treatment protocol |
Six-well plates were coated with retronectin for 16 h, followed by spin at 3000 rpm, 60 min, 4°C with supernatant containing MSCV-IRES-YFP (Ctrl) or MSCV-NMyc-IRES-YFP (NMyc) secreted from GP+E86 cells. HPCs (0.5 × 106/well) from WT, Het, and Hom mice were then seeded on the plates with either Ctrl or NMyc in c-BM media.

Growth protocol |
Lineage-negative BM cells (HPCs) from WT, Het, and Hom mice were separated by the autoMACS. All sorted HPCs were immediately seeded for 48 h in complete BM media (c-BM media) till transduction with NMyc or Ctrl vectors.

Following fragmentation, cRNA were hybridized on Affymetrix Clariom™ S Mouse Array by GeneChip® Hybridization Oven 645 according to the protocols recommended by the manufacturer (Affymetrix, Inc).

GeneChips were scanned using the GeneChip® Scanner 3000 7G (Affymetrix, Inc).
