Title |
Invitrogen yeast protoarray probed with YGL227W WW domain, first experiment |
Sample type |
protein |
Source name |
E.coli overexpression strain
Organism |
Saccharomyces cerevisiae |
Characteristics |
biotinylated WW domain fusion protein
Extracted molecule |
protein |
Extraction protocol |
WW domain probes were purified as fusion proteins from E. coli overexpression strains
Label |
Alexa Fluor 647
Hybridization protocol |
Biotinylated fusion protein probes were applied at a concentration of 50 uM using conditions supplied by Invitrogen. Interactions were visulalized by the application of Alexa Fluor 647-streptavavidin
Scan protocol |
Fluorescent array images were collected for Cy5 with a GenePix 4000A fluorescent scanner and image intensity data were extracted and analyzed with GenePix Pro 3.0 analysis software.
Description |
WW domain MBP-fusion protein expressed and purified from E. coli, and chemically biotinylated with Pierce NHS-LC-LC-biotin
Data processing |
Each protein is printed in duplicate on the array; for each protein, the values of F635_MEAN - B635_MEAN were averaged (VALUE). The arrays are printed in a total of 48 blocks. The mean and standard deviation of signals from each block were calculated. If a protein's VALUE was greater than three standard deviations above the mean for it's parent block, it was called significant.
