At day 7 cells were treated with IL-4 for 48 hours with 10 ng/ml of IL-4.

Growth protocol |
Human ciculating monocytes were isolated using a double gradient of Ficall and Percoll, followed by an additional step with the monocyte isolation Kit II of Miltenyi. Cells were cultured in bacteriological plastic for 7 days in X-Viv0 10 medium supplemented with 1% autologous serum.

Extraction protocol |
RNA was extracted with QIAGEN RNeasy mini kit in accordance with the prescribed protocol provided with the kit. Quality control was performed with Agilent Bioanalyser.
